'O' is considered to be another one letter word in the English language! We see it mostly in poetry and song - "O Christmas tree......" for example. Ad far as meaning, i think that's self-explanatory in the context of the sentence.

I would say just two, I and a, unless of course it takes only one letter words to spell the words we use to describe each letter of the alphabet and of course how many languages your talking about I would assume english but you haven't specified.
